After using them for a couple of days and comparing them to my A161p, I'd say sound signature wise they aren't too different. I think the A161p has faster transients, punchier bass, and a bit more clarity than the R3. Soundstage is bigger on the R3, which also has a bit more air, but weirdly has less sparkle in the treble than the A161p. Comfort-wise I dislike how large and heavy the housing is, which irritates my outer ear after a while and making it hard to get a good seal without Comply tips. The R3 has the beefiest cable I've seen in an IEM. If my A161p (weakest aspect is the frail cable) had that I'd never feel a need to upgrade it.
