I'm torn on what the best to get for about $250 or a little more. I'm an aphile freak! I need good quality headphones, none of that cheap beats by dre stuff.

You're lacking information here.

What setting do you expect to use them in?
Do you currently own an amp/dac?
What sort of genres do you enjoy?
Would you buy used?

+1

I would just cover all the bases and say:
Senn HD25-1 ii
or
ATH-M50 + T50RP

Xonar Essence ST
Every genre except for jazz, classical, etc.
No, I want new.

I'm basically looking for dominant bass that's very smooth, clear and well defined, not muddy and messy; and strong mids, or complimentary mids to bass and treble.

Heya,

For $250, you have a lot of options. The limiting factor for your situation is the sound card though acting as your amplifier. It may make even high impedance headphones play at acceptable volumes, but it won't push the power needed for many of them to reach their potential.

Based on what you said you want (bass), you've got some options.

Sony XB500 (~$50) & Sony XB700 (~$80)
AudioTechnica M50's (~$150)
BeyerDyanimc DT770 Pro 80's (~$200)
Ultrasone Pro 900 (~$300~$400)

I'm biased, but for the money, I'd go with the DT770 Pro's. I probably look/sound like a broken record today with all these Beyer/Bass threads popping up. :p

Very best, :)
