As long as you will not over driven it the answer is no.

The drivers are usually outliving the rest of the headphone don't worry about putting a few hundred hours on it.

Theoretically, yes. Assuming that the drivers have a limited lifetime, any time spent on burn-in will reduce the headphones lifetime.
 
But if burn-in is necessary, the per mil of total lifetime used during burn-in is probably well spent, making the enjoyment of the remaining lifetime much greater.

The reduction of lifetime will be insignificant. Even if you spend 200 hours on burn-in.
 
The reduction of lifetime is purely theoretical, and will probably not be of any importance in real life
