On the description for this cable (Hosatech 3.5 TRS to Dual 1/4), it says it's been designed to connect from the 1/4 to a 3.5mm. Can I connect in reverse: running out the 1/4 to a 3.5mm input? Will it perform optimally or is it not designed for this?
 
Sorry for such a noob question, but I wanted to make sure is the right cable.

Sure it will work.  While a very few interconnects have an added shield that's only connected at the send end,  I don't think that it applies to a complex cable like this. And even then it will still work.
