Most META42's being built and shown on HeadFi seems to be put in fullsize enclosures. If there was no size restrictions for the board can there be any improvements to it? Perhaps the buffer unit implemented onto it.

Well, one thing we see people doing all the time is putting big power caps off-board. A bigger board could end that.

Could probably also fit space for other board mounted components, i.e. like the pots are done today.

Board mounted 1/8", RCA, and 1/4 audio in/out connectors, a power switch, 2.5mm power jack etc. would make assembly cake. No more hookup wire.

And if the board was sized to a specific case, it would be easy to sell the whole works as a kit that could be quickly assembled.

If you don't plan on making something outrageously big like the Gilmore Ultrareg PSU, a board a little under 5"x5" would seem like a good size, so it can still fit into the medium-sized cases. That said, I don't think the Meta42 design should be severed into full-size and mini versions.
