Just wondering if this is possible to do without affecting the sound? I ordered a pair of UE4000's and the ear cushions are non replaceable.

EU4000 is a fishing reel. Could you provide a link to what you are talking about?
Most people who frequent head-fi grow tired of the sound of headphones with non-replaceable ear pads long before the pads wear out.
If you are really motivated, and have too much time, it is possible to rebuild any pad.
