MDF and/or Plexiglass

MDF you can get at the home depot.

Acrylic can be found at most glass suppliers. Just look in the yellow pages. I found it a while back cut to whatever size I wanted for very cheap -- something like $5/sqft.

I'm pretty sure Home Depot sells Acrylic/Polycarbonate sheets (3/16" or whatever it is in thickness) as well.

MDF is the way to go for most designs.
Baltic Birch is an arguably better choice for subwoofer cabinets.
Both can be had at Home Depot or Lowes.

Plexiglass is expensive, and I really don't recommend it. Difficult to work with, flexible, and to get it thick enough that it won't flex, costs a small fortune, and weighs a ton compared to Birch, while MDF is much cheaper and still lighter.

3/4" MDF SHOULD be good for most any speaker application you are building. Some will say 1" is what you need but from my experiance the 3/4" works well.

you can get both from your local lumber yard. Depending on where you are located and how your local yard works the price may be slightly more, the same or even slightly less (usually slightly more though)
however, almost always you will get better service from the lumber yard, it will take less time, and you will get higher quality material.
So, my suggestion would be to get your mdf from a lumber yard and the plexi from a store like home depot.
