I've been searching for a comfortable pair of headphones / headset to wear while gaming and streaming. I have had several gaming headsets over the past couple years and have found most of them to be extremely uncomfortable after a couple hours of use. My search for a comfortable pair of headphones led me to the Beyerdynamic DT 990's 250 Ohms, which I heard are great for gaming. I know very little about audio, so I was hoping someone here could answer a few questions for me.

I own a Behringer Xeynyx X1204 USB that I use for streaming, allowing for easy volume control of my mic, game audio, and music. I usually just plug my headset directly into the mixer. Now I know that any headphones over 100 ohms usually requires a sound card and amp to get a good sound, but does the mixer serve as a substitute for an amp? If not are there any you could recommend that would work with my setup.

I think the DT990s would be a good choice for you; I have the DT770 pro because the wife and I game in the same room and I wanted something to isolate myself as hearing her talk in the room and thorough my headphones on our Vent server at a slight delay was distracting. I moved from HD650s which I found to be way more comfortable than any gaming headphones I've used. The DT770 was even a step above the senns in comfort(oh and I have had my 650s for years so they no longer clamp extremely hard like when new). The beyerdynamic ear cups are big, deep and the pads are supper plush. I went for the DT770 pro vs premium 600 ohm because I read the headband pad on the pro models of the beyerdynamic is a bit thicker and on other gaming headphones this was a major problem for area for me. I also read the pro model was a bit better at isolation.
So I think you are looking at a really good choice for long periods of use, just do some more reading and decide if you want the premiums or the pro model. The premium versions of beyerdynamic headphones are offered in 32, 250 and 600 ohm and the pro versions are only offered in 250 ohm.
I honestly have no idea what the quality of the amp is in your mixer, you could try it and see where you are sound quality wise with the 250 ohm 990s or just get the 32 ohm to be safe. But I would strongly consider looking at a stand alone amp vs buying a gaming soundcard and assuming on that will be card will be good enough. I don't know anyone who runs a high or moderate impedance headphone off any current gaming sound card. Plus it is amazing what you can find in a quality amp for less money than you would spend on a sound card.
If you go the amp route I would be looking at making a choice between the DT990 premium 600 ohm and the DT990 pro 250 ohm. A good amp can power the 600 ohm just fine and most people like those drivers the best in the DT990, so I comes down to either those 600 ohm drivers or the pro headband pad. obviously my other factor of isolation is not relevant to you as the DT990s are semi open.
