Basically I want to know if the Audio Technica Ath M50 will make a really big improvement over my 25$ Sony MDR zx300
Are expensive headphones actually better than the cheap ones?

Yes--of course they will even if you have only a mediocre chain of equipment feeding them.
Two caveats:
#1 "really big improvement" is a very subjective test.
#2 There are better cans available for the money than the M50, although some would still select them because they prefer its other than neutral sound signature and/or they have features/characteristics that some like and prefer.
