Between two what is the less fatiguing during treble and mids reproduction ?
And what give the most unforgiving sound with lower quality tracks ( MP3 or example ).

The HD 600 is a very natural sounding can with warmth to make anything sound good. Not familiar with the 400i.

The HD650 is more relaxed than the HD600. Both the 600 and 650 are more relaxed than the HE-400i too, but it's not a huge difference.

Yes, look at this graph:

1- HD650 is slightly warmer in the bass
2- HD600 is significantly louder between 3000 Hz and 5500 Hz (more forward = less relaxed presentation)
