you must go to MoMA, the Metropolitan Museum and the Guggenheim. while you're doing the Met or the Guggenheim, you should stroll through Central Park.
walking south from the Met through the park, you can see the boat pond, Bethesda Fountain and the Mall. it's a long walk, but if you go all the way down, you'll hit Grand Army Plaza and the Plaza Hotel (now a condo). that's also where the carriage horses pick up, so you could grab a ride on one of those if you're so inclined. touristy, but good fun.
i walk down 5th Ave along the park to work every day from up around the Met, and still love it.
if you fancy a cocktail, you must spend an evening at the Pegu Club on Houston. they handmake all of their proprietary cocktails. the atmosphere and the drinks are unbeatable.
try to get some Korean BBQ in Koreatown (35th St. and north, midtown).
you can see the Statue of Liberty from the Staten Island Ferry, which is free and has a bar. it's a 20 minute (or so) ride each way.
there are so many good restaurants, it's hard to choose. If you're in the Lower East Side, you must stop by Mamoun's Falafel... best.falafel.ever. grab a hot dog or three (and a papaya juice) at either Grey's Papaya or Papaya King.
for lunch on the go, keep an eye out for the Schnitzel & Things truck. usually in midtown (52nd between 6th/7th aves)... but i think they've been venturing downtown lately. i know this is a little too specific... but damn it's good.
