Probably not, but why did you do that? Maybe the clearcoat got mixed up in some of your solder joints and is causing a bad connection.

I took another look at your design. I'm pretty sure everything is in place so it boils down to either a wiring problem or defective component(s).

How may PCB did you fabbed? You should try another but this time no clearcoat spray -- BTW, have any pics of the finished board?

You need to get some pics of the board assembled, (attached is my layout but I airwired the tubes it might give some ideas)

for diy friendly of a pcb board,you will be happy more people can play around it.all in one design is a good idea.
you should setup the volume pot,amp part and power part in one board.

fyi,
there are leader design of a tube hybrid amp.sch design by runhight,pcb design by me and some of headwize members,completed at year 2006.i think this project is a excellent example for you.
