How to match tubes without a tube tester?

Say if I have a bunch of NOS tubes and want to find the match pairs, can I just install the tube, and apply a fixed DC input and measure the output voltage to match them? how does a tube tester do it, does it actually measure plate current or plate voltage?

You should first start with a tube checker rather then a tester. It will check for shorts and other nastiness. The testers are as varied as they are many. Some test the current while others test other parameters and some are more comprehensive then others.
